Do what Ewan McGregor and Charley Boorman did and use a BMW GS R1200 motorcycle. The "Long Way Around" documentary was fantastic and I would recommend to everybody that they see it. Their journey from England through Europe, Asia and finishing in New York is pretty amazing.

Seriously though, I'm an avid motorcyclist and up until a couple years ago used to take a number of solitary bike trips to different places. Things are just too busy now. I've always wanted to take a number of months and see other parts of the world on motorcycle. In a few years I'm going to do just that.
For that purpose I will take either:
One of my beloved KLRs. Simple, rough, uncomfortable at times, but nearly indestructible. Good for pavement, good for dirt. As Peter Egan, the automotive writer once remarked, "At any given time there is a number of KLRs traversing the globe".

Or my Honda Varadero. Six hundred pounds of Honda's interpretation of a big road going dirt bike. V-twin 1000 cc, heavy frame, long suspension travel, and huge fuel capacity. Sold in Europe where they are either traveling dirt roads or performing high speed motorway runs.

Audi A7. Why? It's perfect. 4 doors, "hatchback", nice performance, AWD, breathtaking design, and tons and tons of cabin tech. I'd say CTS-V too, but I'd prefer the smooth ride of an Audi for a long road trip.

it's probably my favorite truck ever, too. 
And nobody mentioned an RV yet? You could basically go on indefinitely on one of those. Dunno if they're fun to drive though, never had the pleasure.
