light wheels

do lighter wheels make for a better DYNO run?

I don't see why they would. They would help in the handling department because it is less unsprung weight.

sure, less rotational mass means the engine has to do less work to accelerate, freeing up some power

yes. do a hub dyno if you're that worried.
