Is this solo-2 ?
For 5 AT, leave it in "L". Most solo-2 courses are short enough to never max out 2nd gear.
Since the TL has a wide turning radius take the hairpins wider; start on the outside as far as possible just like any other car but don't go directly to the apex where the cone is. Take it a little wider or else your rear tire will hit the cone.
As for VSA? I've never autocrossed a car with any type of stability assist so I don't know if it should be left on or turned off.
Oh... and keep both hands on the wheel. You've got a 5AT so just leave it in "L" and don't touch the shifter ever again.

I was curious when I first saw a guy show up in a Lexus GS.
Even running against comparable cars it got smoked badly.
I haven't seen a new TL run yet but I wouldn't count on winning if I were that guy..
