I have found that if you get into your car and have to make a call BEFORE the phone completely connects to the car, you will lose your connectivity and have to re-pair. It's annoying, but it's happened to me a few times. Wait to make your call until you see the phone icons light up in the upper right hand corner of the screen (you know, the signal/bar indicator).

If you try to make a call too soon, it will say "THERE ARE NO PHONES PAIRED WITH THE CAR" and you're stuck.

Of course, you may have a different issue, but this is something to look for.
