Is there a gas mileage difference between front wheel vs SH-AWD
Is there any noticeable gas mileage between a TLX front wheel drive and a TLX with SH-AWD.
I am guessing there is a bit of a difference between a 6 and 4 cylinder engine but what about same engine with different drives?

When you say "noticeable", do you mean other than the numbers published by Acura?

You will get numbers all over the place, depending on how people drive. Most of us didn't get the AWD for mileage, the fun factor did it for me. A few MPG less isn't going to break the bank.
If you baby each car and drive economically, you can get better than published numbers for either.

There are more powertrain losses with driving four wheels instead of two, so yes there is a noticeable difference. You'll probably get 10% or better MPG with FWD.

Also, I think one of the reasons why the SH-AWD's stated mileage is so close to the PAW-S/FWD version is they added the start/stop tech to the SH-AWD cars, which the FWD cars don't have.
